is there a way to manually pull with Kissassist?

The short answer, yes. You can /mqp on to pause it.
Depends on the toon you are running it on vs the toon you're pulling with and the role you have assigned. A bit more info would be helpful to help you.
Are you driving with the SK? Is the SK MT/MA?
Could drive with the SK as MT/MA without using KA at all or use of the roles in KA below

Other than to pause it, not that I know of. The role of "puller" tank would be just that right? If it's a temporary thing, you can pause and then restart after the pull I suppose.
Or change the role to tank and pull with a different toon and let the SK tank it when you return to camp.

I would set the SK as tank and create 2 Hotbutton with "/mqp on" and "/mqp off".
When running off to pull, then push "/mqp on" when coming back to camp push "/mqp off".
This will pause KA while pulling.
I do similar, when pulling with my Bard.

I use the same method Dragonslayer does and it works great for me. I very frequently will also pause everything even when automated pulling and hit return to camp because I see the group getting out of place. Pause on / off is super useful and you can manually pull fine with it and then unpause in camp and let KA or your other automation do its thing. There is probably a more sophisticated way of doing it but I tend to like simple lol.
